1: Does this Coomassie Brilliant Blue staining solution, which does not require destaining, contain methanol and acetic acid? How safe is it?

The formulation contains no methanol or acetic acid—irritating organic solvents—and includes only Coomassie Brilliant Blue G250, a mild acid, and deionized water, with no pungent odor.

Note: The dye solution is mildly acidic and slightly corrosive; always wear disposable gloves and a lab coat throughout the procedure.
2: How long does conventional protein gel staining take, and is a destaining step required?

Under room‑temperature conditions, protein bands can be clearly visualized in just 1–2 minutes, eliminating the need for the tedious steps of fixation, boiling, and destaining.

If deeper staining with higher contrast is desired, the incubation time can be extended to 30 minutes or even overnight.

3: Can the staining solution be recovered and reused after use, and how many times can it be reused at most?

After dyeing is complete, the dye solution can be filtered and recovered into its original reagent bottle; it can be reused up to 2–3 times.

Tip: After multiple reuses, the staining speed may slow down slightly, but it does not reduce the sensitivity of protein detection.
4: If the dye solution develops precipitates or crystallizes at low temperatures, can it still be used normally?

A small amount of sediment in the bottle is normal; shake well to disperse it before use for standard staining.

When crystallization occurs during low-temperature storage, place the entire vial of staining solution in a 37°C water bath to fully dissolve; after cooling, it can be used without affecting the staining results.

5: After staining, the bromophenol blue in the gel turns yellow-green. How can the background color be reduced?

Simply immerse the entire gel in deionized water and let it stand; this will reduce the yellow‑green background caused by bromophenol blue without compromising the protein bands.

6: After staining is complete, no protein bands are visible at all. What could be the cause, and how can this be resolved?

In the vast majority of cases, the protein loading amount is insufficient.

Solution: During electrophoresis, BSA protein was added concurrently as a positive control to identify potential issues in sample loading, electrophoresis, and membrane transfer.
7: After staining, the gel background is relatively high. What methods can be used to reduce background impurities?

1. After electrophoresis, increase the number and duration of distilled water washes to remove impurities from the gel in advance.

2. After staining is complete, immerse the gel in distilled water and allow it to stand at room temperature or at 4°C; overnight incubation at 4°C yields the best destaining results.

8: Which protein gels are compatible? How can the colorimetric sensitivity be enhanced for low-abundance thin-gel samples?

Both denatured and native protein PAGE gels are compatible.
