50 bp DNA Ladder​ is a ready-to-use molecular weight marker composed of ten individual PCR-generated DNA fragments. 

 

It is premixed with loading dye for direct gel loading (recommended volume: 6 µl per well). 

 

The 300 bp reference band is intensified to approximately 100 ng for enhanced visibility, while the remaining nine bands are present at approximately 50 ng each. 

 

The total concentration is approximately 92 ng/µl.

Standard dosage : Load 6 μL per sample

General Recommendations : 3–6 μL per well can both yield clear bands.

6. If the sample well width varies, how should the sample volume be adjusted?

Calculate the volume as 1 μL per 1 mm of sample well width; for wider wells, you may appropriately increase the volume to 3–6 μL, but avoid overloading, which can cause band smearing.

7. Is centrifugation, heating, or vortexing required before sample loading?

It is recommended to centrifuge briefly at low speed. , sink to the bottom to prevent wall adhesion

8. What agarose gel concentration and electrophoresis voltage are recommended?

Gel concentration : 2%–3% agarose gel (best for separating small DNA fragments)

9. How do you visualize the bands after electrophoresis?

After staining with a standard nucleic acid dye, simply visualize the sample under a UV gel imaging system or a UV transilluminator; it is compatible with commonly used dyes such as EB, SYBR Green, and GelRed.

10. Why is the 300-bp band brighter, and could this affect experimental interpretation?

The 300-bp band is a brightly labeled reference marker, used at a higher concentration (100 ng) to facilitate rapid localization and size estimation; the remaining bands exhibit uniform brightness and do not interfere with molecular-weight determination, which is consistent with the intended design.
