Strain Characteristics & Description

E. coliOrigami 2(DE3) Competent Cells contain mutations in the genes encoding thioredoxin reductase (trxB) and glutathione reductase (gor). These mutations disrupt the two primary reducing pathways in the cytoplasm, promoting the formation of correctly folded proteins containing disulfide bonds and thereby enhancing protein solubility.

Kanamycin Sensitivity:​ Unlike the original Origami series, Origami 2 strains are Kanamycin sensitive. This allows for the use of Kanamycin as a selective marker for plasmids, significantly broadening the range of compatible vectors.
Leucine Auxotrophy:​ This strain is a leucine auxotroph (), requiring supplementation with leucine for growth.
Antibiotic Resistance Markers:​ Resistant to Streptomycin and Tetracycline.
Transformation Efficiency:​ Exhibits high transformation efficiency, typically yielding > 10⁷ cfu/µg DNA​ when tested with pUC19 plasmid.
